Can we disable NULL option in LOV's with out making the field mandatory?

Only Yes or No to be present in the drop down and also in backend Database

To disable the null or 'None' option then you will have to make the field Mandatory.
If your intention is to simply replace the 'None' option then you can choose the Label for Null Value and set it to something like 'Unknown' or 'Not Applicable'.
